Quantitative (Phospho)Proteinuntersuchungen in klinischen Gewebeproben

Zusammenfassung

In fast allen Ländern ist das Fixieren von klinischen Gewebeproben mit Formalin und Einbetten in Paraffin zur nachfolgenden histopathologischen Untersuchung ein Standardverfahren, um erkranktes von gesundem Gewebe zu unterscheiden. Proteine ließen sich bisher nur schwer in Formalin-fixierten, Paraffin-eingebetteten (FFPE) Geweben quantitativ untersuchen. Der Grundgedanke unseres Vorhabens ist die Extraktion von intakten Proteinen und phospho-Proteinen aus FFPE Gewebeproben in den klinischen Routinebetrieb zur quantitativen Bestimmung von Biomarkern für die individualisierte Tumortherapie. Das Gesamtziel des Vorhabens ist die funktionelle Signalwegskartierung von Proteinkinasen (z.B. HER-Rezeptoren, PI3K, AKT, ERK) zur Patientenselektion für Therapien mit Signalwegsinhibitoren in soliden Tumoren. Mit unserem national und international bedeutsamen Verfahren werden sich in Zukunft Proteinbiomarker in FFPE-Geweben verlässlich quantifizieren lassen zur Unterstützung Patienten-maßgeschneiderter Therapien ohne den derzeitigen klinischen Ablauf entscheidend ändern zu müssen.
